Methylene Blue
A redox compound showing acute memory effects in healthy volunteers, but unclear benefit in dementia trials.
Evidence category: Clinical study. This describes the source category, not approval or certainty.
Regulatory statement in source: Not separately recorded; do not infer approval from the evidence category.
Methylene blue is a redox-active compound with effects on mitochondrial function and reactive oxygen species. In double-blind, randomized, placebo-controlled trials in healthy volunteers, a single oral dose of methylene blue showed effects on working memory and sustained attention during functional MRI studies, with no negative adverse events reported. A phase II trial examined the same single-dose regimen for effects on cerebral blood flow, functional connectivity, and cognition in healthy aging, mild cognitive impairment, and mild Alzheimer's disease. In Alzheimer's disease trials, oral methylene blue was tested but large studies failed to find clear benefit, partly because blinding was difficult. Phase II malaria trials in children with falciparum malaria showed no drug-related serious adverse events or deaths.
Benefits reported in the imported source
- Shows effects on memory and attention in healthy volunteers during functional MRI testing
- Generally well-tolerated with no serious adverse events in studies
Risks and limits
- Large Phase III trials in Alzheimer's disease failed to show clear benefit
- Blinding difficulties in dementia trials complicate interpretation
- Long-term safety and efficacy in chronic use not established
Source bottom line: Methylene blue shows acute cognitive effects in healthy volunteers but failed to demonstrate clear benefit in Alzheimer's disease trials.

Community and practice descriptions
Accounts of a practice are not evidence of safety or effectiveness. Words such as “standard”, “safe” or “recommended” describe the source account, not verified consensus or this wiki’s advice. Commercial and self-published citations are retained for traceability, not vendor endorsement or independent clinical corroboration.
Historical research outcome: no-verified-description · 2026-10-06
No suitable community description was independently verified in these checks. The entry remains empty; this does not establish that no practice exists elsewhere.

Pharmacokinetic context
Historical half-life estimate: 5.25 hours. Peter, Hongwan, Kupfer & Lauterburg 2000, Eur J Clin Pharmacol, PMID 11214785 — oral/IV crossover in healthy volunteers; the IV and oral arms gave consistent results, unusual for this compound's other pharmacokinetic parameters Population/route context and estimates may differ. This is not a schedule or a verified human dosing rule.
